Anticancer agent 255 is a monocarbonylated curcumin-1,2,3-oxazole conjugate that targets apoptosis mechanisms in cancer cells. It exhibits potent anticancer activity with IC50 values of 8.8 μM and 9.5 μM in prostate cancer cell lines PC-3 and DU-145, respectively, and 6 μM, 10 μM, and 6.4 μM in breast cancer cell lines MCF-7, MDA-MB-231, and 4T1. This compound induces mitochondria-mediated apoptosis, inhibits cell cycle progression, and alters the expression of key proteins by down-regulating the cell proliferation marker PCNA and cell survival proteins, while up-regulating the pro-apoptotic protein Bax and down-regulating the anti-apoptotic protein Bcl-2.
